Question about layering...and locking layers.

I'm having difficulty understanding the "locking" concept in layers...I try to lock layers when working with only a specific area...yet whenever I try to do something TO that layer...the other ones are affected as well...I'm confused.

Also, how does one apply filtering techniques to a layer...like, I can filter specific selections that I made...but once I turn a selection into a layer I seem unable to...any way to fix this?
