You need to link layers together to use those. Select the layer that you want all the others aligned to, then link all the other layers that you want to align with whatever actions you pick
Or, on a shape layer (or any layer with a vector mask) select more than one path
The first set of buttons are for alignment. The 2nd set of button are for distribution, which requires 3 or more layers linked or selected paths

What you need do it activate these is first have an object on the canvas, the select all (Crtl+a) then press V. These options will the be active.
